Phospholipase C( PLC)活性が亢進していることを報告してきた。本研究は,冠攣縮性狭心症のメカニズムを解明するために,CSA 患者および対照被験者の皮膚線維芽細胞から人工多能性幹細胞(iPSC)を樹立し,更に,iPSCを平滑筋細胞(SMC)に分化誘導させ,CSA の病態に関与する細胞内Ca2+ 流入の分子機構を調べた。\n【方法と結果】細胞内遊離Ca2+ 濃度([Ca2+]i)はFura-2 AM を使用し測定した。アセチルコリン(ACh)10-4M 誘導性の[Ca2+]i は,対照iPSC 由来SMC と比しCSA iPSC 由来SMC で優位に上昇した(53±10 versus 129±24 nM, p <0.05)。前処理ジルチアゼム10-4M では,対照iPSC 由来SMC では42±1nM(p <0.05),CSA iPSC 由来SMC では72±10 nM(p <0.01)に抑制された。ウェスタンブロット解析の結果,TRPC3 およびPLC-δ1 は差はなかったが,p122RhoGAP/DLC-1 およびTRPC6 の蛋白発現は,対照iPSC 由来SMC と比しCSA iPSC 由来SMC で,それぞれ2.60±0.8および1.68±0.28倍増加していた(p <0.05)。PLC 活性は,対照iPSC 由来SMC に比しCSA iPSC 由来SMC において有意に高かった(3.12±1.28倍,p <0.05)。\n【結論】DLC-1 およびTRPC6 タンパク質の過剰発現とPLC 活性の増強は,CSA 患者における細胞内[Ca2+]i
